MicroRNAs (miRNAs) are noncoding RNA molecules around 22 nucleotides in length that are primarily recognized for their role in post-transcriptional gene regulation. Most commonly, miRNAs silence messenger RNA targets by base-pairing with complimentary sequence in the 3’ UTR of the target transcript. Though general rules about this miRNA:target association have been defined, the base pairing between miRNA and target is typically imperfect, and therefore miRNA binding sites are difficult to accurately predict computationally. The Turner lab focuses on performing experimental studies to validate computationally predicted miRNA targets. The work presented here is focused on validating predicted targets of mir-72, a miRNA shown to be associated with Cystic Fibrosis, and demonstrates a general pipeline that can be universally applied to aid in validating the predicted targets of all miRNAs. It is our hope that in doing so, the experimentally validated lists of miRNA targets will aid in honing the efforts of medical research physicians and scientists, as well as pharmaceutical companies, to identify biomarkers and targets that will aid in both diagnosis and treatment of disease.
