The protein encoded by the p250GAP mRNA is known to be associated with nerve plasticity in dendrites. The p250GAP mRNA translation has been shown to be regulated by the microRNA miR-132p, this regulation being dependent on the presence of the fragile X mental retardation protein (FMRP). p250GAP mRNA has two key regions that demonstrate potential to be associated with FMRP. The first is a G-rich region that has the potential to form a G quadruplex, a secondary structure known to be bound with high affinity and specificity by FMRP. The second region contains a poly U stretch, another sequence proposed to be recognized by FMRP. In this study we have used biophysical techniques such as H<sup>1</sup> NMR spectroscopy, CD spectroscopy, UV-Vis spectroscopy, and Native PAGE to analyze the secondary structures of these two regions in p250GAP mRNA, as well as their interactions with FMRP.
