Polyketide natural products are privileged motifs of diverse structural and functional complexity and provide important sources of novel pharmaceutical agents. During polyketide biosynthesis, the acyltransferase (AT) domain selects specific acyl-CoA donors and loads acyl units onto a downstream acyl carrier protein (ACP). To understand this elusive biological process, we present our efforts to develop novel tools to study AT domains in polyketide synthases (PKSs), including AT-specific fluorescent probes and mechanistic crosslinkers. This toolkit may facilitate profiling of AT domain activity in whole cells and may enable the interrogation of transient AT-ACP protein-protein interactions.
