Biopharmaceutical drugs are often formulated at high protein concentrations.  These highly concentrated formulations alter the drug’s properties and increase viscosities making characterization difficult.  Infrared spectroscopy is one method that can still be used to determine concentration and structural descriptions of concentrated protein formulations.  We are developing new infrared methods to measure the structures, dynamics and functions of nearly identical biopharmaceutical protein samples.  We have recently used infrared spectroscopy to characterize both the structures of drug proteins as well as their associated water molecules.  We will present descriptions of structural changes, induced by high concentration, in model polymers and proteins including the NIST reference monoclonal antibody.
