Lingzhiol is a structurally unprecedented meroterpenoid isolated recently from mushroom <i>Ganoderma lucidum</i> widely used in traditional Chinese medicine under the name <i>ling-zhi</i>. It was found to possess potent and selective inhibitory activity towards p-Smad3, a transcription protein implicated in renal fibrosis. We have proposed a plausible biosynthesis of lingzhiol, which has inspired us to develop a concise and flexible synthetic route to this natural product.
