Batrachotoxin (BTX) was first isolated in 1965 from the venom of the Columbian poison arrow frog, <i>Phyllobates</i> <i>aurotaenia</i>. Its unprecedented structure contains a tetracyclic steroidal core, a seven-membered oxazapane ring, and a bridging hemiketal. It is the most potent small molecule agonist of voltage-gated sodium ion channels. To date, an enantioselective total synthesis has not been reported. A convergent synthesis will provide sufficient material for biological testing, and enable the preparation of strategically modified analogues. A summary of our efforts towards the enantioselective total synthesis of BTX will be presented.
