Trisulfide bonds are formed from sulfur addition into protein disulfide linkages and have been reported in monoclonal antibodies and other proteins. Trisulfides are an important product quality attribute (PQA) in antibody drug conjugate (ADC) molecules and may also be an important PQA in non-ADC molecules. We have observed varying trisulfide levels in some molecules even using the same media and process conditions; therefore, an effort was taken to improve our understanding of the mechanism and to identify levers to control trisulfides and improve process robustness. Experiments were conducted to better understand the mechanism of trisulfide formation and to identify media components that act as levers during cell culture to minimize trisulfide formation.
