Multimodal, or mixed-mode, chromatography combines several modes of interaction between ligands and target molecules in commercial downstream bioprocessing.   Specifically, mixed-mode anion exchange resins take advantage of the ionic and hydrophobic target molecule properties for downstream purification as well as possibly providing viral clearance. Viral clearance by downstream unit operations like chromatography is critical for establishing the safety of biotech medicines produced by mammalian cells.  Preliminary research by our group suggests that there are additive effects of the hydrophobic and electrostatic interactions between viruses and mixed mode chromatography.  While there are several ways to measure viral electrostatic properties, there is very little relative or quantitative data available for viral hydrophobicity.  An assay to measure viral hydrophobicity would aid in further explorations of viral interactions with mixed mode chromatography.  In this project we explored the utility of two different methodologies, hydrophobic interaction chromatography and a fluorescent thermal stability assay, in measuring viral hydrophobicity, starting with bacteriophage models.  Specifically, with these assays we were able to ascertain the relative hydrophobic values of three different bacteriophage models for mammalian viruses in comparison to known protein standards. <br/>
