C60 is a carbon nanomaterial that is widely used in chemical industries ranging from electronics to medicine. Given its widespread use, human exposure to C60 is probable and therefore the potential for its adverse effects must be investigated. Previous work on C60 toxicity reports various contradicting facts especially regarding the toxic doses of C60. In this study, we treated mouse liver cells (Hepa-V) with a wide range of C60 doses (ppb to ppm in 7.5% bovine serum albumin) for 24 hours and monitored cell viability, ATP levels, reactive oxygen species (ROS) generation, and mitochondrial membrane potential. Cell viability was determined using the MTT (soluble salt) assay and ATP production was measured with a luciferin/luciferase luminescence assay. Mitochondrial membrane potential and ROS generation were determined using JC-10 and Green cell ROX, respectively, and analyzed using flow cytometry. The results show that low doses of C60 do not have significant effects on cell viability, ATP production, ROS production and mitochondrial membrane potential. However, higher C60 doses have negative effects on cells showing increased cell death and ROS production and decreased ATP production. These results provide information useful in establishing permitted C60 exposure levels.
