The LC-MS analyses of different types of enzyme catalyzed reaction in biological samples were performed using AB SCIEX Triple Quad 6500 coupled with WATERS ACQUITY UPLC I CLASS System available in our High Throughput Analytical Chemistry Center. Examples of enzyme converted metabolites of nucleoside triphosphate are determined and confirmed by LC-MS(Q3). Similar method developed also used for analysis of enzymatic reaction of amino acid substrates in cell culture media. Conditions of column, different mobile phases, UV absorption wavelength, mass spec methodology, flow rate and temperature are examined and optimized. The developed method can effectively reduce the running time of injections and successfully provide a resource to track the content of substrates in different enzyme reactions.
