Hydrogen sulfide has recently been discovered as a significant gasotransmitter, joining the ranks of CO and NO.1 Some roles of H2S in the body include relaxing vascular smooth muscle, inducing hibernation, inhibiting insulin signaling, regulating inflammation and dilating blood vessels.2 Developments in our understanding of the biological roles of H2S have led to a surge in research focused on new methods of H2S donation. This therapeutic potential for H2S may provide a route to develop pharmacologically-active H2S donors useful in both research and medicine. A significant limitation of current H2S donors, however, is that it is difficult to control when H2S is released. Based on this need, we report here new methods for H2S donation in which an external stimulus can be used to trigger H2S release from the donor scaffolds. We report the synthesis and characterization of these compounds as well as studies confirming H2S donation in solution upon donor activation. We envision that these new types of H2S donors could be useful research tools that enable controlled H2S release in different biological environments. References 1Wang, R.
