Solid-state-NMR-based structural studies of membrane proteins and complex biomaterials require high resolution to obtain site-specific information and high sensitivity to detect low-abundance species. High-resolution SSNMR can be achieved by a combination of high magnetic fields, multidimensional correlation techniques, and pulse sequences that detect a subset of all signals at a time. Enhanced sensitivity can be achieved by, among other things, dynamic nuclear polarization. I will show our recent progress in developing new experiments with enhanced resolution and sensitivity to elucidate the structural basis of ion conduction in a membrane-bound ion channel and to understand the mechanism of protein-carbohydrate binding in plant cell walls.
