Replacing the phosphodiester backbone of RNA with positively charged guanidinium linkages produces ribonucleic guanidine (RNG). RNG has been shown to overcome electrostatic repulsion and bind double-stranded DNA in a triplex with high affinity. Ribonucleotide monomers with the ability to form guanidinium linkages have been synthesized for the generation of ribooligonucleotides with guanidinium linkages (RNGs). Here we describe an efficient method for the synthesis of 5'-amino and 3',5'-amino ribonucleoside analogues with appropriate protecting groups to be compatible with the solid-phase synthesis of cytidyl and uridyl RNG oligonucleotides.
