Extensive research efforts have been made over the last two decades on developing highly sensitive and selective biosensing platforms in conjunction with gold nanoparticles for a wide range of applications. In this talk, we will demonstrate some of our latest efforts made on the use of biofunctionalized gold nanoparticles (NPs), surface sandwich configurations and surface enzyme reactions to enhance both the sensitivity and selectivity of bioaffinity sensors for proteins which can be further applied in biomedical diagnostics. The first part of my talk contains the development of a new surface-based sandwich assay with biofunctionalized Au NPs for the SPR detection of a cardiac failure biomarker, B-type natriuretic peptide (BNP) at an attomolar concentration level in undiluted serum samples.1 In addition, another DNA aptamer-antibody surface sandwich assay was developed for the selective detection of alpha-1 antitrypsin2 and Tau proteins in both serum and plasma samples, which could potentially be employed for the diagnosis of Alzheimer’s disease. The second part of my talk contains the combined use of biofunctionalized Au nanoparticles and surface enzyme reactions for creating electrochemical biosensing platforms for femtomolar concentrations of H5N1 viral proteins in serum samples.3 For example, the selective and sensitive detection of H5N1 can be voltammetrically determined via the electrocatalytic reaction of the substrate, 4-aminophenylphosphate with the surface sandwich complexes of H5N1-aptamer/H5N1/antiH5N1-alkaline phosphatase on a Au NP modified screen printed carbon electrodes.
