Bispecific antibodies provide an opportunity to target two different antigens with a single therapeutic.  Applications range from combining two drug activities into one molecule to novel therapeutic mechanisms. However, this advancement in antibody engineering brings new process challenges.  This talk will focus on the assembly steps of bispecific antibodies from separately expressed halves. Solution conditions including pH, temperature and reducing agent all play a role in assembly kinetics and require optimization to maximize assembly efficiency. Case studies will be presented on the assembly of bispecific antibodies from different half-antibodies using knob-hole technology.
