Ruthenium(II) polypyridyl complexes with distorted octahedral geometry represent a new class of photodynamic therapy agents whose mechanism of action involve covalent binding with DNA. Herein, we will discuss synthesis, characterization, ligand exchange properties, electrochemical properties, photophysical properties, and DNA interaction properties of a series of sterically strained Ru(II) polypyridyl complexes and Ru(II)-Pt(II) bimetallic complexes. Design criteria for development of tumor selective PDT agents will be discussed.
