Azadirachta indica, commonly known as Neem tree, is one of the oldest medicinal plants used in the world. The Neem tree grow mostly in India. Among its applications are properties associated with the immune system, dermatological, anti-cancer, diabetes, and possible insecticide. The objective of this research is to analyze the effects of extracts of Neem leaf in the cytochrome P450 system of Chinese Hamster Ovarian cells (CHO cells). CHO cells were cultured and sub cultured. Dried leaves and undried leaves extracts were prepared. Cells were treated with different concentrations of the extracts: 100 mg/L, 300 mg/L and 3000 mg/L. Microsomes were prepared and the following assays were performed: Bradford, Cytochrome P450 content, p-Nitrophenol Hydroxylase, Erythromycin-N-demethylase (ERND), and 7-Ethoxyresorufin deethylase (EROD). Protein content, following Bradford assay, showed a higher concentration for treated cells with dry and undried leaves extracts (300 mg/L). ERND activity decreased for dried and undried leaves extracts, except for cells treated with dried leaf extract of 300 mg/L. EROD enzyme activity decreased for all extracts concentrations. p-Nitrophenol hydroxylase showed a higher activity for the extract of the dried leaf of 3000 mg/L. Cells treated with the extract of the dried and undried leaf of 300 mg/L increased the concentration of cytochrome P450.Components of the extracts will be studied using the method of HPLC and the effects of these components will be analyzed on the activity of the isoenzymes. The results will be validated by performing statistical analysis methodology
