Employing a key ring closing metathesis, we have synthesized a new class of heterocycle containing sulfur, phosphorus, and nitrogen known as a thiol phosphonamidate. This concise four-step synthesis allows for diversification at several key positions with modification of the starting materials. We herein report on the progress of two amino-acid based diversification towards an external and internal diversification at the nitrogen. Ultimately, these diversifications will provide several options to synthesize dozens of new thiol phosphonamidates to create a library of unique derivatives which we believe will display potent biological activity due to repeating motifs of common pharmaceuticals.<br/>
