Surface-enhanced Raman scattering (SERS) nanoparticles offer important advantages over traditional imaging probes, including ultrahigh detection sensitivity, spectroscopic multiplexing, label-free detection, multivalent targeting, favorable pharmacokinetics, and the potential for multifunctionality and multimodality. There are major opportunities to exploit these features for image-guided surgery and for ultrasensitive detection and tracking of metastatic tumor cells and stem cells. Its development has experienced a number of milestones including the initial discovery in 197, the reports of single-molecule and single-nanoparticle SERS in 1997, and the demonstration of in vivo SERS in 2008. The current wave of activities has focused on cellular tagging, image-guided surgery, and single-molecule/single-particle spectroscopy. The next breakthrough is likely the development of SERS nanopartricles and spectroscopic devices for clinical human applications.
