The overall goal of this research project is to synthesize a molecule that could potentially inhibit type IV prepilin peptidase (TFPP), an enzyme critical for pili formation in <i>Vibrio cholera</i>. The targeted molecule is hypothesized to inhibit TFPP through the combined use of a reduced amide functionality along with a unique substrate sequence based on the known substrate preference of this enzyme. The project focus up to this point has been on optimizing the synthetic pathways necessary for the synthesis of this inhibitor, as the methods reported in the literature give low yields and products that are challenging to purify.  We have been conducting a series of trial reactions on a small scale, helping conserve materials while still allowing multiple runs in order to vary reaction conditions and determine their influence. The results of these trials will be presented.  In the future, the completed molecule will be subjected to biological testing in an autoagglutination assay in order to determine its effectiveness as an inhibitor of TFPP.
