In the last decade, ruthenium (II) polypyridyl complexes have been getting a lot of attention. They have been found to have a wide range of applications such as protein binding and DNA intercalation. In this study, a series of ruthenium (II) polypyridyl complexes with ferrocenyl thiosemicarbazones has been prepared and their interaction with DNA has been investigated. The compounds studied have been synthesized from the reaction of Ru(polypyridine)2Cl2 with the thiosemicarbazone ligand under refluxing conditions. The complexes were characterized using elemental analyses and various spectroscopic methods. The interaction of the complexes with DNA has been investigated by fluorescence and absorption titration. The binding constants of the studied complexes have been found to be of the order of 104 M-1. The interaction of the complexes with DNA was also investigated by cyclic voltammetry and molecular modeling. One of the complexes, [Ru(bipyridine)2(FcTSC)]2+ showed a decrease in the oxidation potential with an increase in the DNA concentration via cyclic voltammetry. The molecular modeling has been used to determine the binding constants for the complexes when they bind to duplex and quadruplex DNA. For [Ru(bipyridine)2(FcTSC)]2+ the calculated binding constant for interaction with duplex DNA was 24.5 μM and for quadruplex DNA, 31.72 μM. This indicates that the complex favors the quadruplex DNA, which is implicated in tumorigenesis. Additional studies carried out include computing the partition coefficient and viscometric studies.
