The Archaeal species Sulfolobus solfataricus is believed to contain two genes that code for the PurF protein, which is necessary to catalyze the first reaction of the purine biosynthesis pathway. The goal of this project is to successfully clone both genes into pTWIN1 plasmids and to eventually express and characterize the proteins. Optimization of the PCR reaction, digests, and ligation conditions has been required. Progress towards cloning both genes will be reported.
