Several DNA and RNA sequences have been correlated to the occurrence of cancer. Accurate measurement of these molecules is therefore critical in biosensing applications. We have designed a microfluidic sensor that relies on the thermodynamic stability of fully hybridized duplexes and the sensitivity of electrochemical methods to measure DNA and RNA levels indirectly. Hybridization of target sequences is reported via downstream detection of displaced redox-labeled oligonucleotides. Optimization of the sensor design, as evaluated by UV-visible spectroscopy and cyclic voltammetry, will be presented.
