Human plasma is used as a manufacturing feedstock for a number of licensed IgG therapeutics in both immunity and infectious disease. Part of the mechanism of action for some of these therapies relies on the engagement of Fc gamma receptors (FcgRs) on immune cells, which in turn elicit effector functions such as antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis. Leveraging FcgRs as chromatographic affinity ligands, we were able to enrich in high affinity subclasses and glycoforms using human plasma derived IgG as a load material. These elution peaks were additionally evaluated for their potentiation of both Natural Killer cell degranulation (ADCC surrogate) as well as Monocyte phagocytosis in cell-based assays. This work demonstrates the use of FcgR-based chromatography to directly impact IgG effector function and illustrates the potential use of this technology to develop new therapeutics from plasma IgG.
