Nanospheres have received much attention in recent years for potential controlled-release, drug vehicles. Here we created triggerable nanospheres which contain a remote-actuatable material inside, notably magnetic nanoparticle(s) responsive to external magnetic field. The mechanisms involved in the observed switchable drug release were investigated. Remotely applied magnetic field induces the associated drug molecules to be released from the nanosphere on-demand. The rate of diffusional drug release is controllable by the degree and duration of magnetic field as well as the chemistry and structure of the capsule materials involved. The penetration of tumors and crossing of the blood-brain barrier (BBB) by the drug delivery cargo are also important for the cancer and the central nervous system (CNS) treatment. With the magnetic nanocapsules fabricated, we demonstrate, in vitro and in vivo, that the magnetic vector can be utilized to guide the drug cargo to the intended location and release the drug by applied magnetic field.
