Oxidation reactions are rarely used in the synthesis of active pharmaceutical ingredients (APIs) for a variety of reasons including the use of halogenated solvents, toxic stoichiometric oxidants, high energy reagents, and resulting hazardous byproducts. From this perspective, oxygen is an attractive and atom economical choice of oxidant. However, aerobic oxidation has found little use in API synthesis, largely due to safety issues associated with combining oxygen rich atmospheres and flammable organic solvents in multipurpose batch reaction vessels typically found in pharmaceutical manufacturing plants. Recent advances in continuous flow process development have enabled a new paradigm for API synthesis where aerobic oxidation reactions can be done safely with an appropriately designed reactor.<br/><br/>This presentation will describe the development of a continuous flow aerobic oxidation to produce a strained ketone en route to an API starting material. Our approach to safety for aerobic oxidation, screening techniques and the resulting scaled up flow process will be discussed.
