Over the past several years, the development of “Greener” synthetic routes has played an increasingly significant role in early discovery research within Pfizer with the recognition that such chemistry improvements can facilitate scale-up, and thus expedite the progression of a compound through the discovery cascade. In the early days, the availability of practical guidance tools like solvent and reagent guides have influenced a chemist’s decision-making and have led to the implementation of more sophisticated techniques such as high-throughput reaction screening on micro scale, and the increased uptake of biotransformation. An overview of the strategic impact of these tools on several projects within our current Oncology portfolio will be discussed, focusing on synthetic route optimization at an early stage to not only increase the rate of analogue production but also enable the large scale production of key molecules for <i>in vivo</i> studies.
