<i>Gynostemma pentaphylla </i>has been consumed as a popular functional tea worldwide. Its saponins were responsible for several beneficial effects including anti-obesity, however the underlying mechanisms and active components remain unclear. In this study, a novel saponin (JS) was isolated from tetraploid and characterized for its chemical structure. JS was aslo investigated for its inhibitory effect on adipocyte differentiation and the potential molecular mechanisms using 3T3-L1 murine pre-adipocytes. JS treatment down-regulated the expression of peroxisome proliferator-activated receptor g (PPARγ) and CCAAT/enhancer-binding protein a (C/EBPα) at both mRNA and protein levels. Detailed analysis proved that the inhibition was primarily limited to the early stage of adipocyte differentiation, where JS decreased the DNA-binding activity of C/EBPβ by blocking its phosphorylation. Furthermore, JS treatment increased expression of β-catenin and inhibited glycogen synthase kinase 3β activity modulating Wnt/β-catenin pathway. The anti-adipogenic effects of JS were significantly attenuated in β-catenin siRNA-transfected cells compared with the control.
