In this presentation we will discuss some of the recent advances in two-dimensional liquid chromatography (2D-LC). Some of these have been theoretical developments, but most have been advances in the practice of 2D-LC. Specifically, we will highlight some of the progress made in column selection and system optimization on the more theoretical side. On the practical side, we will highlight some of the significant developments in methodology including work on hybrid modes of 2D-LC such as multiple heartcutting and selective comprehensive 2D separations. We will also review some of the work that has been focused on the valve interface between dimensions and its effects on 2D-LC performance, including effects on second dimension column stability and effects on detection sensitivity at the second dimension detector. Finally, we will briefly discuss some recent applications of 2D-LC in a variety of application areas that highlight the tremendous capabilities of modern 2D-LC for solving challenging separation problems.<br/>In the second part of the talk, we will discuss in a bit more detail recent work in our laboratory related to the optimization of the second dimension in particular. Specifically, we will discuss the impact of connecting tubing on total system pressure drop under conditions needed for very fast second dimension separations. We will also discuss the impact of the volume and composition of fractions transferred from the first dimension column on the performance of each second dimension separation, with an eye toward both chromatographic resolution and detection sensitivity. In our view the future of 2D-LC is very bright as the base of experienced users continues to grow and learn how to efficiently solve challenging separation problems using modern methodology and instrumentation.
