Despite the recent breakthroughs, current cancer immunotherapy is often confronted with challenges such as low overall response rates. Vaccines can potentiate antitumor immunity via synergistic pathways with other immunotherapeutics. However, molecular vaccines are suboptimally efficacious due to unfavorable delivery to lymph nodes (LNs), where antitumor immunity is coordinated. We developed albumin-binding vaccine (AlbiVax), which leveraged endogenous albumin as nanocarriers for efficient LN-targeted vaccine delivery, as demonstrated by positron emission tomography, γ-counting, and light sheet fluorescence microscopy. AlbiVax ameliorated systemic dissemination and toxicity. AlbiVax induced potent immunostimulation, elevated production of IgG2a antibody, expanded the repertoires of peripheral antigen-specific CD8+ T cells and tumor infiltrating CD8+ T cells, and upregulated PD-1 expression on these T cells. In mice, AlbiVax dramatically inhibited progression of target lymphoblostoma for both prophylactics and therapy; AlbiVax also potentiated PD-1 inhibitor for treatment of established primary and metastatic melanoma. AlbiVax represents a promising potent and safe molecular vaccine.
