The α,α’-disubstituted medium-sized cyclic ethers are abundant as structural cores in biologically important natural products. However, despite of a number of methods developed for medium-sized cyclic ether synthesis, the stereoselective synthesis of 8-membered cyclic ethers is still challenging because of enthalpic and entropic barriers. The cationic gold(I) complexes have emerged as particularly active catalysts for the regio- and stereoselective functionalization of C=C bonds with carbon and heteroatom nucleophiles to form carbocycles and heterocycles. Herein, we disclose our exploration of the gold(I)-catalyzed intramolecular alkoxylation in a stereoselective formal synthesis of (+)-laurencin.
