Despite its reputation as a foul-smelling and toxic pollutant, H2S is a vital biological signaling agent, and it is of interest as a therapeutic for a variety of diseases and conditions. This work focuses on developing polymers for the sustained and controlled delivery of hydrogen sulfide (H2S). Small molecule H2S donors are compounds that can be triggered to decompose with release of H2S in response to a biological trigger. H2S donors used in this work including S-aroylthiooximes and N-thiocarboxyanhydrides, which are appended onto amphiphilic polymer frameworks to form H2S-releasing assemblies, including micelles and vesicles. H2S release rates can be controlled by changing the type of assembly as well as the mobility of the H2S-releasing hydrophobic block. These polymers are relevant as both biological tools and as potential therapeutics, including in cancer, wound-healing, and cardiovascular disease.
