Chloride concentrations along the endosomal pathway have been shown to increase with a decrease in pH to maintain the electrochemical gradient on the membrane. However, our understanding of the role of Cl– in other cellular compartments is limited. In order to understand how Cl– impacts other cellular functions, we have developed a series of fluorescent Cl– sensors, functionalized for ratiometric sensing in vivo. Current work is focused on preliminary studies in aqueous solutions and in vivo measurements of [Cl–]. Future work includes mapping the Cl– concentrations across the cell, and investigating how changes in external stimuli affect Cl– concentrations. We anticipate that these probes have the potential to become unique tools for delving into disease states with a Cl– imbalance.
