Quinones are key structural components in many natural products, and therapeutic drugs impacting many disease conditions, viz antimicrobial, antiparasitic, anti-tumor, inhibition of PGE2 biosynthesis and anti-cardiovascular disease. Our research group has been investigating anthraquinones as kinase inhibitors which revealed a series of quinone analogs with excellent growth inhibition potency for refractive breast tumors and showing high selectivity for inhibition of the PIM and Casein kinases. While pursuing synthetic studies on new quinone analogues we discovered a novel method for the synthesis of binaphthyltetraones using Diels-Alder reaction of substituted (E)-((1-ethoxybuta-1,3-dien-1-yl)oxy)trimethylsilane dienes and benzoquinone dienophile which on further rearrangement and followed by deprotection of the phenolic -OH gave the binaphthyltetraone analogues. The bioactivity of these compounds is being investigated. The results of a complete study of this new synthetic method is presented.
