There is currently a significant need for designed RNA molecules in basic biochemical research as well as areas such as drug design.  Synthesis of RNA oligonucleotides is a solid phase method similar to DNA oligonucleotide synthesis.  Synthesis of RNA phosphoramidite building blocks is very similar to well-established DNA phosphoramidite synthesis methods.  However, due to the additional hydroxyl group in the 2’ position of RNA, the phosphoramidite synthesis results in lower regioselectivity creating a large amount of byproduct.  In an attempt to improve yields of the proper RNA phosphoramidite regioisomer, a derivative of the traditional dimethoxytrityl protecting group was made. Compatibility studies with the new dimethoxytrityl group and the conventional solid phase synthesis method are being conducted to test the dimethoxytrityl derivative in comparison to the conventional 5’-protecting group in the synthesis of RNA. Presented here is the synthesis of dimethoxytrityl derivative and results from the compatibility studies with RNA solid phase synthesis.<br/>
