Single protein spectroscopy has long been a goal of analytical chemists. Myoglobin is a particularly interesting protein due to its biological importance and the sensitivity of the UV-Soret band formed by conformational changes of the heme-core. To investigate how the conformation of heme changes when bound to a surface, myoglobin will be mixed with ND solution to form a ND-myoglobin complex. The effect of the ND on the conformation of the protein will be observed using UV-Vis spectroscopy. Quantification of the amount of protein bound to ND will be determined using a Bradford assay. This ND-myoglobin complex will be disturbed with changed in pH and temperature.
