We have been investigating the thermodynamics of unusual DNA conformations that form from the human telomere sequence; in particular, the C-rich strand (CCCTAA)x, which forms the so called i-motif under slightly acidic conditions. These conformations are stabilized by unusual hydrogen bonding schemes and possess stacked C:CH+ base pairs adjoined by TTA loops. By exploring the both heat of protonation and the osmotic properties of the i-motif for the human telomere sequence (CCCTAA)4, we hope to better understand both the formation and stability of the i-motif. We have been carrying out isothermal titration calorimetric and osmotic pressure analysis. Here, we report the progress on these findings.
