This presentation will describe studies of triphenylmethane dyes that change color and become fluorescent upon binding to oligomers of peptides derived from the β-amyloid peptide, Aβ. We recently reported X-ray crystallographic structures of trimers and higher-order oligomers derived from Aβ. The three-fold symmetrical hydrophobic surfaces of these trimers display three phenylalanine residues and complement triphenylmethane dyes, such as crystal violet. Triphenylmethane dyes bind to stabilized homologues of the trimers, changing color from violet to blue and fluorescing red when irradiated with green light. Ongoing efforts to elucidate binding interactions and to create triphenylmethane dyes with modified selectivity and response will be described. Applications of the dyes to identifying novel ligands for Aβ-derived oligomers will be discussed.
