Since the restriction in the use of the brominated flame-retardant (polybrominated diphenyl ethers, PBDEs), the consumption of OPFRs has increased. Organophosphate triesters are used as additive flame retardants and plasticisers in polymers, paints, synthetic rubber, varnishes, lacquers, and hydraulic fluids.<br/>To monitor the exposure of these OPFRs in California population, our laboratory has developed an analytical method for the determination of dialkyl and diaryl phosphates (DAPs), which are metabolites of organophosphate triesters (OPFRs) in human urine. Target DAPs includes bis (1, 3-dichloro-2-propyl) phosphate (BDCPP), diphenyl phosphate, or diphenyl hydrogen phosphate (DPP), bis (2-chloroethyl) phosphate (BCEP), and bis (1-chloro-2-propyl) phosphate (BCPP). Sample preparation is based on solid phase extraction using a weak anion exchange sorbent. The quantitation is achieved using reversed phase liquid chromatography–negative electrospray ionization tandem mass spectrometry (LC–ESI-MS/MS). Method accuracy (% recovery) and analytical precision or inter-day imprecision (CV%) was calculated using quality control material at three different levels and analyzed over a period of 20 days. This presentation will cover sample preparation, mass spectrometer optimization, liquid chromatographic conditions optimization, matrix effects, quality control material preparation and validation.
