Carbazole and its derivatives have been widely detected and reported throughout the environment in soil, river and lake sediments. Exposure to these compounds shows an adverse health impacts, and the exposure levels can be evaluated via biomonitoring. This presentation addresses the method development and validation for the analysis of carbazole and 12 halogenated carbazoles in human blood samples. Small volume of human blood sample (100 µL) was employed for the analysis. Proteins in human blood samples were removed by precipitation through either using MgSO<sub>4</sub> or using a mixture of formic acid and acetone. The analytes were then extracted into hexane /ethyl acetate or hexane / dichloromethane depending on protein precipitation methods. The extracts were dried under gentle stream of N<sub>2</sub> and then re-dissolved in toluene. Sample analysis was performed using gas-chromatography coupled with tandem mass spectrometer in an electron impact mode. The developed method has been validated and demonstrated negligible matrix effect, low detection limits in the range of 0.05 – 0.5 ng/mL with a satisfactory recovery. Further investigations on lowering the detection limits and the determination of other halogenated carbazoles are underway.
