Quinoline-4-carboxylic acids have been found to have a variety of pharmacological activities, including potent anti-viral and anti-cancer. One such compound, Brequinar, is known to inhibit dihydroorotate dehydrogenase, an enzyme in the process of pyrimidine synthesis. The venerable Pfitzinger reaction is often the method of choice for preparation of quinoline-4-carboxylic acids. We are employing 5- and 6- bromo-, chloro- and fluoro-isatins, propio-aryl compounds, and the Suzuki reaction to prepare a series of 2-substituted 3-methylquinoline-4-carboxylic acids in the quest for enhanced pharmacological utility. These compounds will be bioassayed for anti-viral and anti-cancer activity.
