Split-protein biosensors have been developed in the past for the sequence-specific detection of DNA and covalent DNA modification. SEquence Enabled Reassembly (SEER) of split reporter proteins has proven to be a robust and versatile approach to the detection of DNA in its native, double-stranded state. The demonstrated modular nature of DNA binding and signaling domains in previously reported SEER systems indicates that this approach to biosensor development could be suitable for research at the undergraduate level. As such, we have begun to develop biosensors using previously unexplored DNA binding domains. The current work describes biosensors based on an inactivated variant of a CRISPR associated protein complex (dCas9), as well as the high mobility group box 1 protein (HMGB1).
