Fluorescence experiments investigating the interactions between cytochrome c and labeled lipid vesicles containing 50 mole% 1,2-diacylphospitidylglycerol lipids [10:0, 12:0, 14:0, 16:0, 18:1(△9 cis)] with 1,2-dioleoyl-<i>sn</i>-glycero-3-phosphocholine (DOPC) in pH 7.4 buffered solutions are reported. Fluorescence titration experiments show that cytochrome c has a similar affinity for absorption onto the surfaces of the 50 mole% PG lipid vesicles. Fluorescence recovery experiments reveal the incomplete desorption of cytochrome c upon the addition greater than a 10X excess of unlabeled PG vesicles. The extent of desorption is related to the length of the PG lipid’s acyl chain and is independent of temperature, albeit over a narrow temperature range. The differences observed in the extent of cytochrome c desorption are attributed to the relative contributions of the two reported binding motifs reported for cytochrome c to acidic lipid vesicles.
