Natural products have been, and continue to be a rich source of bioactive compounds that are used in the treatment of various diseases. Reniochalistatin E (IC<sub>50</sub> of 4.9 mM towards myeloma & 9.7 mM towards gastric cancers), and lagunamide C (IC<sub>50</sub> ranging from 2.1 to 24.4 nM against numerous cancerous cell lines) are two recently isolated natural products possessing potent anticancer activity. As such, they are of synthetic and biological interested to our research program in the search, synthesis, and development of new anticancer agents. To be presented is the total synthesis of reniochalstatin E and analogs, biological evaluation and current applications into possible therapy strategies. In addition, work towards the total synthesis and structural elucidation of lagunamide C will be presented.
