A series of trinulcear ruthenium complexes were synthesized. The compounds are based on the N2,N2,N4,N4,N6,N6-hexa(pyridine-2-yl)-1,3,5-triazine-2,4,6-triamine (TRA) ligand core and are formulated as [TRA{(diimine)2Ru}3]+ where diimine = 2,2’-bipyridine 1, 1,10-phenathroline 2, 2-(pyridine-2-yl)benzo[d]thiazole (pbt) 3, 11-aminopteridino[6,7-f][1,10]phenanthrolin-13(12H)-one (pterin), 4, pteridino[6,7-f][1,10]phenanthroline-11,13(10H, 12H)-dione (allox), 5. The biophysical properties of the complexes were investigated by following their interactions with DNA via absorption titrations, viscometry and cyclic voltammetry. The antibacterial profiles of the complexes against common microbes including S. aureus, S. typhimurium, E. faecalis, P. aeruginosa, K. pneumonae, B. cereus and C. albicans., were studied by the disk diffusion method and by using a microdilution assay based on the dye, resazurin. Compound 2 showed the best activity particularly against P. aeruginosa (MIC = 62.30 μM) and B. cereus (MIC = 31.15 μM). Complex 1 show little activity. Consequently, a theoretical comparison between this complex and 2, was carried out by molecular docking with bacterial topoisomerase (topo IV) and DNA gyrase B and calculating their respective binding energies.In addition, the nature of the interactions in the docked poses will be discussed.
