High-throughput screening (HTS) plays an important role for drug discovery. Rapid advancement in automation, computer and assay technology has led to the generation of huge amount of research data by screening laboratories. However, HTS data sharing among the screening community was largely missing in the past. To fill up the gap, PubChem BioAssay was set up in 2004 as an open access database by the National Center for Biotechnology Information (NCBI) of NIH. PubChem BioAssay now serves as the largest public data repository for biological results from small molecule and RNAi high-throughput screenings (HTS). With a decade long development, PubChem BioAssay has grown rapidly in data content, archival capacity, informatics utility and user bases. The development efforts of PubChem, the contributions from the community, and the bioassay data content will be described and discussed.
