Englerin A is a natural product with potent and selective anti-cancer activity against renal cell carcinoma (RCC). There is a need for new, selective RCC drugs: current drugs targeting RCC are not effective as single agents, fail to provide long-term solutions, and carry strong adverse side effects. The current total syntheses of Englerin A are lengthy and not economically viable. Progress towards an efficient 9-step synthesis of a structural analog of Englerin A has been made. The progress features a desmethyl analog of the polycyclic guaiane core of Englerin A, the synthesis of which includes a novel Michael addition and a radical cyclization using divalent samarium iodide.
