Microbes are detected by the pathogen-associated molecular patterns (PAMPs) through specific host pattern recognition receptors (PRR). Nucleotide binding oligomerization domain-containing protein 2 (NOD2, CARD15, IBD1) is an intracellular PRR that recognizes fragments of bacterial cell wall.  Nod2 is important to human biology, as when it is mutated, it loses the ability to respond properly to bacterial cell wall fragments.  In order to determine the mechanisms of misactivation in the Nod2 Crohn’s mutants, a combination of carbohydrate synthesis, microbiology and cell biology were used to probe the mechanisms of activation by bacterial cell wall fragments.  This talk will focus on the unexpected finding that Nod2 is an unstable protein and the mechanisms that the cell uses to stabilize this protein, including molecular chaperones, ligand binding and post-translational modifications. <br/>
