Deoxypodophyllotoxin (DPT) is a natural product with known anticancer activity. DPT is structurally similar to podophyllotoxin, which is used to produce the FDA approved chemotherapeutics etoposide and teniposide. Like other natural products, DPT suffers from supply problems being available only in small amounts from natural sources. A comprehensive analysis of the biological activity of DPT is impossible until DPT can be accessed through a synthetic pathway. A 7-step racemic synthesis of DPT is now complete. Progress toward enantiopure DPT through a novel asymmetric styryl-alkynyl-Diels-Alder reaction has been made. The final enantioselective pathway will inexpensively provide an abundance of DPT.
