Phenotypic stability plays a critical role in maintaining the productivity of a cell line during cell culture processes.  To ensure phenotypic stability is maintained, top clones are generally grown for a defined number of generations. This mimics the doublings needed for production scale operations from vial thaw to harvest.  During such a stability study, the top three clones of a high-performance NS0 cell line were found to have been unstable, with a titer loss of up to 77% after 60 population doublings.  We found the cause of the cell line instability to be the choice of medium for cell growth.  A detailed study of the medium components indicated the instability was a result of one raw material.  In addition, we generated data supporting the hypothesis and demonstrated that the cells maintain stability in a modified media for up to 80 population doublings.  We also determined that intracellular staining was useful for predicting stability for NS0 cells, which could shorten phenotypic stability study timelines.<br/>
